I know, people do that Everyone has different associations and experiences and beliefs about different names, and no one name will ever be agreed on by everybody. I just think you totally ignore them and pick a name you and your OH love and think fits your little one Even if they hate the name initially, once they get to know your little baby those ideas go out the window, and then your family couldn't imagine him/her with a different name
Personally, I dislike everyone asking us about names right now. My DH has made it very clear he doesn't want to talk about names until the baby is here, which to me is stressful! We've been arguing about it (finally both did some compromising), so I hate feeling that pressure from people asking what we're going to name him! It's an emotional topic for me right now, haha, so I feel like I have to hold back when people ask! lol

Should Hector Lombard drop to welterweight after debut loss at UFC 149?

Hector Lombard came into UFC 149 riding a long win streak and an unstoppable hype train. He walked out of UFC 149 with a decision loss and a disappointed boss.

"It wasn't the fight I was expecting," UFC president Dana White admitted. "It just wasn't what I thought it was going to be. Whether it was Boetsch or Lombard winning, I was expecting a real war. I thought this thing was going to be a war."

It was far from an exciting bout, as Lombard, known for his aggression and knockouts, showed little aggressiveness in the bout. White wondered if Octagon jitters got the best of Lombard, and also threw out the idea that middleweight is too big of a weight class.

"Lombard maybe should fight at 170," White said. "He made 185 easily. He's short and wide, but I think he could make 170."

In a Q and A before the UFC 149 weigh-ins, middleweight Michael Bisping made jokes about Lombard's height.

"I bumped into Hector Lombard in the elevator, and I thought someone was playing a practical joke. I was like, 'Who is this dwarf?' I think he's in the wrong weight class. I think he's in the 125 weight class."

At 5-foot-9, Lombard is one of the shortest middleweights in the division. Most 185ers in the UFC are at least six-foot. Champion Anderson Silva is 6-foot-2. Though fighters have overcome height and reach differences in bouts -- think Pat Barry knocking out Stefan Struve -- there is no reason for Lombard to come into the fight at a disadvantage if he is comfortable dropping down.

Foster parent contemplating domestic adoption - Adoption.com ...

Hi Everyone,

I have been a foster parent for 3 and a half years. I adopted my first placement, and since then I've only had 2 other placements, both of which were temporary. I enjoy providing foster care and want to continue being a foster parent. However, my daughter is 4 now and I feel very ready to adopt again.

I chose foster care originally because of financial reasons. Mainly - adopting from the state is very inexpensive and Social Services pays for the adoption. I am still not rich by any means, but I am in a better financial situation than I was when I started looking into adoption.

So, I am now contemplating the idea of domestic infant adoption, and I have plenty of questions. First of all, while I have been able to foster/adopt as a single parent, I have felt slightly discriminated against. I got my placements when all "couples" homes were full or not taking placements. I've had the least placements of my group in DSS. So, I'm just wondering how far that discrimination runs - have any of you felt like you had less of a chance of adopting, because of your family dynamics?

I want to ask opinions on adoption agencies as well, but I'm unclear about whether you need to use a local agency or you can use one out of state? Either way, are you presented to birth mothers only with your local area, or anywhere within the US?

With that said, I'd love to hear from some of you about the agencies you used and whether you would recommend them or not. I know some agencies seem to work better with singles than others do.

Any and all advice is welcome. I'm not brand new to adoption, but I am brand new to domestic adoption and would love to learn as much as I can!

Syrian forces bombard Damascus, fight rages in Aleppo

BEIRUT/AMMAN (Reuters) - Syrian forces bombarded three districts of Damascus with helicopter gunships on Sunday, witnesses said, clawing back territory from rebels a week after the fighters launched what they called a final battle for the capital.

Fighting also raged around the main intelligence headquarters in Syria's biggest city, Aleppo -- the country's main commercial and industrial hub -- and in Deir al-Zor on the Euprhates river, the largest city in the east.

Rebels said they had captured a third border crossing with Turkey on Sunday, Bab al-Salam north of Aleppo, while Iraqi officials said Syrian forces had regained control of one of two border crossings seized by rebels on the frontier with Iraq.

The helicopter bombardments in Damascus and Deir al-Zor showed President Bashar al-Assad's determination to regain control after a bomb killed four members of his high command in the gravest blow in the 16-month-old revolt.

Rebels were driven from Mezze, the diplomatic district of Damascus, residents and opposition activists said, and elite Fourth Division troops were besieging the northern neighbourhood of Barzeh, one of three northern areas hit by helicopter fire.

The fourth division is run by Assad's younger brother, Maher al-Assad, 41, who is widely seen as the muscle maintaining the Assad family's four decades of Alawite minority rule.

His role has become more crucial since Assad's defence and intelligence ministers, a top general and his powerful brother-in-law were killed by the bomb on Wednesday, part of a "Damascus volcano" by rebels seeking to turn the tables in a revolt inspired by Arab Spring uprisings in Tunisia, Libya and Egypt.

Assad has not spoken in public since the bombing. Diplomats and opposition sources said government forces were focusing on strategic centres, with one Western diplomat comparing Assad to a doctor "abandoning the patient's limbs to save the organs".

Syrian state television quoted a media source denying that helicopters had fired on the capital. "The situation in Damascus is normal, but the security forces are pursuing the remnants of the terrorists in some streets," it said.

Assad's forces, who also pushed into a rebel-held district in the northerly commercial hub of Aleppo on Saturday, targeted pockets of lightly armed rebels, who moved about the streets on foot and attacked security installations and roadblocks.

Residents said the sound of shelling in the capital was so intense at dusk that they were unable to distinguish it from the traditional cannon blast marking the end of the daily fast for the Muslim holy month of Ramadan.

Opposition activists said late on Saturday that helicopters had fired rockets into a neighbourhood near the southerly Sayida Zeinab district, causing dozens of casualties. They did not have any other details.

"In Damascus, people continue to search desperately for safety," the International Committee of the Red Cross (ICRC) said in a statement in Geneva.

"Humanitarian needs are growing as the situation in the city worsens and as large numbers of people flee their neighbourhoods in search of safe haven. The ICRC and the Syrian Arab Red Crescent have intensified their response to the situation."

The Syrian Observatory for Human Rights, an opposition group that monitors the violence, said 180 people, including 48 troops, had been killed across Syria on Saturday. Many of them died in the province of Homs, epicentre of the uprising.

Most shops in Damascus were closed and there was only light traffic - although more than in the past few days. Some police checkpoints, abandoned earlier in the week, were manned again.

Many petrol stations were closed, having run out of fuel, and those that were open had huge lines of cars waiting to fill up. Residents reported long queues at bakeries.

FLIGHT FROM ALEPPO

A bloody crackdown on what began as a peaceful revolt has increasingly become an armed conflict between an establishment dominated by Assad's Alawite minority, an offshoot of Shi'ite Islam, and rebels drawn largely from the Sunni majority.

U.N. Secretary General Ban Ki-moon said he was sending his peacekeeping chief Herve Ladsous and top military adviser General Babacar Gaye to Syria to assess the situation.

Opposition activists in Aleppo said hundreds of families were fleeing residential areas on Saturday after the military swept into the Saladin district, which had been in rebel hands for two days.

Fighting was also reported in the densely-populated, poor neighbourhood of al-Sakhour.

"The sound of bombardment has been non-stop since last night. For the first time we feel Aleppo has turned into a battle zone," a housewife, who declined to be named, said by phone from the city.

REBEL BORDER CROSSING RAID

On the Iraqi-Syrian border, Iraqi security and border officials said Syrian forces had reasserted control over the Yarubiya crossing point on the Syrian side of the frontier, briefly seized by rebels on Saturday.

Syrian opposition activists said several towns in Syria's Kurdish northeast had passed - without a fight - into local hands in recent days as central authority eroded.

The surge in violence has trapped millions of Syrians, turned sections of Damascus into ghost areas, and sent tens of thousands of refugees fleeing to neighbouring Lebanon.

The U.N. Security Council has approved a 30-day extension for a ceasefire observer mission, but Ban has recommended changing its focus to pursuing prospects for a political solution - effectively accepting there is no truce to monitor.

Diplomats said only half of the 300 unarmed observers would be needed for Ban's suggested plan, and several monitors were seen departing from Damascus on Saturday.

Speaking two days after Russia and China vetoed a resolution to impose U.N. sanctions on Assad's government, Ban called on he Security Council to "redouble efforts to forge a united way forward and exercise its collective responsibility".

"The Syrian government has manifestly failed to protect civilians and the international community has collective responsibility to live up to the U.N. Charter and act on its principles," he said.

Regional and Western powers have voiced concern the conflict might become a full-blown sectarian war that could spill across borders. But Assad's opponents remain outgunned and divided.

French Foreign Minister Laurent Fabius, speaking after contacts with the head of the Arab League and Qatar's prime minister, said all three agreed that it was time for Syria's fractured opposition to prepare to take charge of the country.

"We would like to see the rapid formation of a provisional government representing the diversity of Syrian society," said Fabius. Syria's main political opposition group, the Syrian National Council, operating in exile, has so far failed to unite Assad's disparate foes on a united political platform.

A Golden Age for Lithium-Air EV Batteries?

Lithium-ion batteries have become standard for portable electronics because they don?t require a lot of power to run. However, using Li-ion batteries for bigger purposes, like powering an electric car, does not bode as well. Many of the newest electric vehicles can run for only 100 miles (if that) before needing a recharge.

Among the many alternative technologies researchers are investigating is lithium-air, a way to build batteries with a much higher energy density than Li-ion. This week in the journal Science, a group of chemists at the University of St Andrews in Scotland led by Peter Bruce reports on a prototype battery that can more efficiently use oxygen in the atmosphere as a source of power. All it takes is a little gold.

The idea of lithium air is that it could gather the oxygen it requires from the atmosphere, which would allow the battery to be lighter because it wouldn?t need to store heavy metal oxides. The reactions inside the battery would form lithium peroxide while the battery was expending energy, and, in theory, the opposite reaction would happen while the battery was charging, releasing oxygen back out into the air.

But in most prototypes so far, which used carbon-based electrodes, the battery didn?t stand up to repeated discharging and charging cycles, due in part to unwanted reactions happening on the side. That degredation meant the battery wasn?t fully rechargeable. But by making the electrodes out of gold (and adding in some dimethyl sulfoxide), Bruce and colleagues got rid of these side reactions almost entirely. Once all the lithium ions have been used up, all it takes is a flick of a switch to reverse the reaction and make them available once again. Even after 100 charging cycles, the battery retained 95 percent capacity.

Bruce and his colleagues acknowledge that car batteries with electrodes of pure gold don?t make much economic sense. However, he puts forth the idea that it?s possible to take a carbon electrode and thinly coat it in gold to achieve the same results.

Sony Vaio T13 review - Bonnie Cha - Product Reviews - AllThingsD

If you?re shopping for a new laptop, you might find the term ?Ultrabook? popping up a lot during your research. An Ultrabook is thinner and lighter than a regular laptop, but it doesn?t sacrifice such features as a full keyboard, and it uses a different kind of memory instead of traditional hard drives to offer speed and good battery life, so you can use it as your main computer for everyday tasks like browsing the Web and working on documents. After a trend started by Apple?s MacBook Air, Windows PC makers such as HP, Dell and Samsung offer them, and now Sony has joined the fray.

Over the past week, I?ve been testing the Sony Vaio T13, the company?s first Ultrabook, and what makes it stand out from the pack is its affordable price tag. The cost of Ultrabooks still hovers in the $1,000 range but the Vaio T series starts at $770. By comparison, the similarly featured Dell XPS 13 costs $999 and up, while the 13-inch MacBook Air starts at $1,199.

For the price, the Vaio T13 offers Intel?s latest Ivy Bridge processors, a plethora of ports and four gigabytes of memory. To keep costs down, there are some tradeoffs, such as a lower resolution screen, but overall, I found the Vaio T13 to be a speedy machine. It?s a good value to be sure, but there are some downsides, such as a subpar keyboard and loud fan, that holds it back from excellent.

The Vaio T13 measures 12.7 inches wide, 0.71 inch thick and weighs 3.54 pounds, so it?s heavier than a number of ultrabooks on the market. For example, the Dell XPS 13 is 2.99 pounds and the MacBook Air weighs 2.96 pounds. That said, I didn?t find the extra weight to be a huge problem. I was able to carry it around in my backpack without it weighing me down.

Also, while it?s thicker than the Air, I appreciated the selection of ports available on the Vaio. This includes an Ethernet jack, two USB ports, HDMI and VGA ports if you want to connect to a TV or monitor and a Memory Stick Pro Duo/SD card slot.

The Vaio T has a 13.3-inch display with a 1,366 by 768 pixel resolution. Colors looked a bit dull, and pictures and videos weren?t overly bright or sharp. The lower resolution also means you have to scroll a bit more to see what?s onscreen, but you do get a decent-size touchpad that supports multitouch gestures to help you navigate various pages and switch between tasks. It worked well in my tests as I was able to return to the desktop by swiping down on the touchpad with four fingers and move between Web pages and photos by swiping left or right with three fingers.

But I didn?t like the Ultrabook?s keyboard. There?s plenty of spacing between the keys, so it doesn?t feel cramped. But the buttons are stiff and squishy, so I really had to punch at the keys. It?s something I got used to after a while, but it led to a lot of mistakes and frustration at the beginning. I also wish the keyboard was backlit for easier typing in darker rooms.

The $799 Vaio T13 I reviewed is powered by an Intel Core i5 processor and features a combined 500GB hard drive and 32GB solid-state drive. (You can configure the ultrabook with a faster i7 processor and up to 8GB of memory.) A number of Ultrabooks feature only solid-state drives ? or flash memory ? that improve speed and battery life but limit storage capacity. They are also more expensive, which is why Sony used this hybrid option.

Even so, I found the Vaio T13 to be a responsive computer and you get the bonus of extra storage capacity. It took the Ultrabook 29 seconds to boot up from a cold start and seven seconds to wake up from sleep mode. The MacBook Air started up in 15 seconds and awoke in five seconds, so there isn?t that much of a difference in the latter test.

At one time, I had at least a dozen tabs open in my Web browser, streamed a YouTube video and downloaded an album from the Google Play Web store while working on a Word document. The Vaio T13 was able to handle all those tasks without slowing down. However, its fan does tend to make a lot of noise. It?s annoying, but I suppose it is doing its job, as the Ultrabook never got warm, even when I was viewing entire TV shows on Hulu.

Sony estimates the Vaio T13?s battery life to be around seven hours and 30 minutes with power-saving mode on. In my tests, where I turned off all power-saving features and left Wi-Fi on, set the display to full brightness and looped a music playlist while running an email application in the background, the Vaio delivered four hours and 20 minutes of battery life.

The battery performance is middle-of-the-road compared to some of the other Ultrabooks we?ve tested at AllThingsD, with the Dell XPS 13 being the worst at under four hours and the Lenovo IdeaPad U300s being the best at five hours. With more normal usage, I expect you should get about an additional hour of battery life from the Vaio T13.

The Vaio runs Windows 7 and comes preloaded with a starter edition of Microsoft Office 2010, which offers basic versions of Word and Excel, and a 30-day trial of Kaspersky Internet Security. There are also various Sony apps, such as the company?s Music Unlimited store and PlayMemories multimedia gallery ? all of which you can access from a pull-down menu at the top of the desktop screen.

For those on a budget, the Sony Vaio T13 is certainly worth a consideration. It?s fast and offers a good amount storage. But if possible, I?d definitely recommend going into a store and trying out the keyboard to see if it?s to your liking.

Consumers are more likely to select products located in the horizontal center of a display and may not make the best choices as a result, according to a new study in the Journal of Consumer Research.

"A close investigation of visual attention reveals that consumers do not accurately recall their choice process. Our findings emphasize the relationship between horizontal location, attention, and choice," write authors A. Selin Atalay (HEC Paris), H. Onur Bodur (Concordia University), and Dina Rasolofoarison (Aston Business School).

Many products are arranged horizontally. For example, rows of snack bars in a vending machine, bottles displaying the beer selection in a bar, or jars of peanut butter on a supermarket shelf. How does this influence which option a consumer will choose?

Using eye-tracking devices, the authors investigated how location influences choices for products as varied as vitamins, meal replacement bars, and energy drinks. Consumers had a tendency to increase their visual focus on the central option in the final five seconds prior to a decision and this determined which option they would choose. Consumers did not accurately recall their choice process and were not aware of any conscious visual focus.

Another study in a retail environment demonstrated that the centrally located item within a product category is chosen more often, even when it is not placed in the center of the shelf or visual field. Consumers would make better choices if they were aware that their attention usually focuses on the center.

"In the context of low involvement choice between frequently purchased products, when choosing between unfamiliar yet equivalent brands, the visual search process and consumer choice are biased toward centrally located options. Being unaware that our attention is focused on the center can lead to poor choices," the authors conclude.

Obesity plus low vitamin D may add up to a greater risk of diabetes

ScienceDaily (July 17, 2012) ? The combination of obesity and vitamin D deficiency may put people at even greater risk of insulin resistance than either factor alone, according to new research from the Drexel University School of Public Health recently published early online in the journal Diabetes Care. Insulin resistance is a major risk factor for Type 2 diabetes, a condition that affects 25.6 million adults and is the seventh leading cause of death in the United States.

"Vitamin D insufficiency and obesity are individual risk factors for insulin resistance and diabetes," said lead author Shaum Kabadi, a doctoral candidate in epidemiology at Drexel. "Our results suggest that the combination of these two factors increases the odds of insulin resistance to an even greater degree than would have been expected based on their individual contributions."

In the study, obese individuals who had healthy levels of vitamin D had insulin resistance almost 20 times more often than the overall study population. But in obese individuals whose serum vitamin D was low, insulin resistance was much higher: about 32 times more common than the average.

Senior author Dr. Longjian Liu, an associate professor in the School of Public Health, noted, "It's not clear whether obesity itself causes a low vitamin D level or if it's the other way around."

Vitamin D is stored in adipose fat tissues, making it unavailable for the body to use; as a result, people who are overweight are already more likely to have low levels of serum vitamin D. Vitamin D deficiency is associated with multiple health conditions including diabetes, cardiovascular diseases including stroke, depression, dementia and other conditions.

Kabadi, Liu and co-author Dr. Brian Lee, an assistant professor in the School of Public Health, analyzed data on serum vitamin D levels and indicators of insulin resistance and diabetes from 5,806 respondents to a major national health survey, the National Health and Nutrition Examination Survey (NHANES). This was the first study of the association between vitamin D and diabetes risk for obese patients using a large, nationally representative sample of adults. The survey reported data from individuals at a single point in time and was therefore unable to determine whether there is a cause-and-effect relationship among vitamin D, obesity and insulin resistance.

Further studies could indicate whether vitamin D supplements are effective at reducing the risk of insulin resistance and diabetes in obese individuals. If so, the authors noted that it would be an inexpensive and practical prevention strategy compared to the difficulty involved in healthy weight loss.

Liu said that vitamin D supplements may be useful for people who are overweight or obese to help control diabetes, but he cautioned that too much vitamin D can cause side effects such as weakness and fatigue. In addition, vitamin D deficiency is just one of many known risk factors associated with insulin resistance and diabetes. "Therefore, to control diabetes, we need to have comprehensive prevention strategies. For example, physical activity may be the most cost-effective way to control weight and subsequently to control conditions including diabetes and heart disease," Liu said.

Open-Access of UK-Funded Science Papers Will Start in 2013

From April 2013, science papers must be made free to access within six months of publication if they come from work paid for by one of the United Kingdom?s seven government-funded grant agencies, the research councils, which together spend about ?2.8 billion (US$4.4 billion) each year on research.

The policy, announced this morning by the agencies? umbrella body Research Councils UK (RCUK), makes clear that researchers should shun science journals that don?t allow authors to follow this mandate.

Also this morning, the UK government formally welcomed the Finch report into open access (which it had commissioned). Its response makes clear that RCUK?s new policy is the driving force for change.

RCUK hasn?t said how it will sanction those who don?t comply. (Astrid Wissenberg, who chairs the RCUK Impact Group, tells Nature that it will be looking to push to ?75% compliance over a number of years?). But if it does rigorously enforce the policy, that will mark a dramatic shift for scientists, publishers and universities ? perhaps the most significant change on the ground since Britain?s science minister David Willetts began discussing how to improve access to research papers more than a year ago.

Open ambition

RCUK released a draft version of the policy in March, and the final version makes no important changes, as the agencies had received mostly supportive comments, says an RCUK spokesperson. In essence, it is similar to announcements from the UK?s Wellcome Trust, a major biomedical research charity. But the research councils? move will be more influential, as Wellcome spends only ?600 million each year. Work funded in part by the research councils counts, so the policy includes overseas researchers collaborating with British scientists.

The research councils have said since 2006 that they want research to made free as soon as possible after publication. The difference today is that they are firmly stating the six-month maximum delay and, most importantly, are announcing how they will take money out of research grants to pay for open access.

Science journals have two ways of complying with the policy. They can allow the final peer-reviewed version of a paper to be put into an online repository within six months. Alternatively, publishers may charge authors to make research papers open-access up front.

In the United Kingdom in 2010, authors paid for immediate open-access publication for some 5% of papers (known as ?gold? open access), and another 35% were put into repositories after first being behind a paywall (?green? open access) ? except that some of those were not the final peer-reviewed version of the paper. (The proportions also vary between disciplines, as you can see from this chart.)

For ?gold? open access, RCUK will pay institutions an annual block grant to support the charges. If government doesn?t give RCUK any more cash, the money required will come from existing grant funding; it?s been previously estimated at some 1?1.5% of research budgets. In turn, RCUK expects that institutions will set up and manage their own publication funds. That might mean that universities and researchers will begin to discuss where they can afford to publish.

The changes?brought on by?these new communication and distribution channels are in many ways revolutionary, and they?re causing disruptions akin to those of past revolutions.

Beijing, July 14: Chinese police has said that a second investigation into the death of leading dissident Li Wangyang showed that he had committed suicide.

Authorities in the country?s Hunan province said that the conclusion was made in consultation with forensic experts and experienced investigators.

According to the BBC, Wangyang was found dead in his hospital room in the central Chinese city of Shaoyang on June 6.

Wangyang spent more than 22 years in jail after taking part in the Tiananmen Square protests of 1989, and his death sparked off protests in Hong Kong.

Following his death, a senior Hong Kong official said he doubted the nearly deaf and blind activist could hang himself.

According to the report, Chinese authorities claim he had committed suicide while under police guard.

But Wangyang? friends had suggested that the suicide seemed to be staged and accused authorities of rushing to cremate the activist?s body following an autopsy. (ANI)

Discovery opens door to attacking biofilms that cause chronic infections

A clever new imaging technique discovered at the University of California, Berkeley, reveals a possible plan of attack for many bacterial diseases, such as cholera, lung infections in cystic fibrosis patients and even chronic sinusitis, that form biofilms that make them resistant to antibiotics.

By devising a new fluorescent labeling strategy and employing super-resolution light microscopy, the researchers were able to examine the structure of sticky plaques called bacterial biofilms that make these infections so tenacious. They also identified genetic targets for potential drugs that could break up the bacterial community and expose the bugs to the killing power of antibiotics.

"Eventually, we want to make these bugs homeless," said lead researcher Veysel Berk, a postdoctoral fellow in the Department of Physics and the California Institute for Quantitative Biosciences (QB3) at UC Berkeley.

Berk and his coauthors, including Nobel laureate Steven Chu, report their findings in the July 13 issue of the journal Science.

"In their natural habitat, 99.9 percent of all bacteria live as a community and attach to surfaces as biofilms; according to the National Institutes of Health, 80 percent of all infections in humans are related to biofilms," Berk said.

The researchers were able to employ new techniques that allowed them to zoom into a street-level view of these biofilms, where they learned "how they grow from a single cell and come together to form rooms and whole buildings," Berk said. "Now, we can come up with a logical approach to discovering how to take down their building, or prevent them from forming the building itself."

Combining super-resolution microscopy with the technique Berk developed, which allows continuous labeling of growing and dividing cells in culture, biologists in many fields will be able to record stop-motion video of "how bacteria build their castles," he said.

In a statement about the paper, Chu said, "Advances in single molecule methods based on optical microscopy have made stunning progress in the past two decades. In particular, 'super-resolution' imaging methods enabled researchers to image structures with unprecedented detail. Veysel Berk led a team that applied these optical methods to continuously image live biofilms on length scales on tens of nanometers to tens of microns. Before this work, these bacterial communities were largely studied in terms of their average composition, appearance and bulk biochemistry. By introducing a new in vivo fluorescence tagging strategy, the molecular and architectural roles of three specific matrix proteins and the extraracellular polysaccharides of a growing V. cholerae bioflim were visualized as series of three-dimensional images. This work has led to new insights into the development of these complex structures and will no doubt pave the way to new approaches to fighting infectious disease and also bacteriological applications in environmental and industrial settings."

Bacteria are not loners

The popular view of bacteria is that they are free-living organisms easily kept in check by antibiotics, Berk said. But scientists now realize that bacteria spend most of their lives in colonies or biofilms, even in the human body. While single bacteria may be susceptible to antibiotics, the films can be 1,000 times more resistant and most can only be removed surgically.

Implants, such as pacemakers, stents and artificial joints, occasionally become infected by bacteria that form biofilms. These biofilm sites periodically shed bacteria ? adventurers, Berk calls them ? which can ignite acute infections and fever. While antibiotics can knock out these free-swimming bacteria and temporally calm down the infection, the biofilm remains untouched. The only permanent solution is removal of the biofilm-coated device and replacement with a new sterilized implant.

A permanent bacterial biofilm in the sinuses can ignite an immune response leading to chronic sinus infections, with symptoms including fever and cold-like symptoms. So far, the most effective treatment is to surgically remove the affected tissue.

Bacteria also form permanent, mostly lifelong, biofilms in the mucus-filled lungs of cystic fibrosis patients and are responsible for the chronic lung infections that lead to early death. Although long-lasting antibiotic treatment helps, it cannot eradicate the infection completely.

To study a biofilm formed by cholera bacteria, Berk built his own super-resolution microscope in the basement of UC Berkeley's Stanley Hall based on a 2007 design by coauthor Xiaowei Zhuang, Chu's former post-doctoral student who is now a professor at Harvard University. To actually see these cells as they divided to form "castles," Berk devised a new technique called continuous immunostaining that allowed him to track four separate target molecules by means of four separate fluorescent dyes.

He discovered that, over a period of about six hours, a single bacterium laid down a protein glue to attach itself to a surface, then divided into daughter cells while secreting a protein that cements the daughters cells to one another. The daughters continued to divide until they formed a cluster ? like a brick and mortar building ? at which point the bacteria secreted a protein that, along with sugar molecules, encased the cluster like the shell of a building.

The clusters are separated by microchannels that may allow nutrients in and waste out, Berk said.

"If we can find a drug to get rid of the glue protein, we can move the building as a whole. Or if we can get rid of the cement protein, we can dissolve everything and collapse the building, providing antibiotic access," Berk said. "These can be targets for site-specific, antibiotic medicines in the future."

Super-resolution microscopy: painting with light

Berk is a biologist trained in physics and optics with expertise in imaging the structures of proteins: He was part of a team that a few years ago determined the atomic-scale structures of the ribosome, the cellular machine that translates genetic message into a finished protein.

He suspected that powerful new super-resolution light microscopy could reveal the unknown structure of biofilms. Super-resolution microscopy obtains 10 times better resolution than standard light microscopy ? 20 instead of 200 nanometers ? by highlighting only part of the image at a time using photo-switchable probes and compiling thousands of images into a single snapshot. The process is much like painting with light ? shining a flashlight beam on a dark scene while leaving the camera shutter open. Each snapshot may take a few minutes to compile, but for slow cellular growth, that's quick enough to obtain a stop-action movie.

The problem was how to label the cells with fluorescent dyes to continuously monitor their growth and division. Normally, biologists attach primary antibodies to cells, then flood the cells with fluorescent dye attached to a secondary antibody that latches onto the primary. They then flush away the excess dye, shine light on the dyed cells and photograph the fluorescence.

Berk suspected that a critically balanced concentration of fluorescent stain ? low enough to prevent background, but high enough to have efficient staining ? would work just as well and eliminate the need to flush out excess dye for fear it would create a background glow.

"The classical approach is first staining, then destaining, then taking only a single snapshot," Berk said. "We found a way to do staining and keep all the fluorescent probes inside the solution while we do the imaging, so we can continuously monitor everything, starting from a single cell all the way to a mature biofilm. Instead of one snapshot, we are recording a whole movie."

"It was a very simple, cool idea, but everyone thought it was crazy," he said. "Yes, it was crazy, but it worked."
